Wet granulation, dry granulation (roll compaction) and direct blending are the three most common granulation processes for solid dosage form production in the pharmaceutical industry. Given the importance of granulation in the development of oral dosage forms— most fine pharmaceutical compounds need granulation to improve their flow capacity and processing properties until tabulation— and the widespread use of the technique in the industry.
The method of applying a liquid solution to powders involves the rubbing with a granulating solvent of a mixture of dry primary powder particles. The fluid contains an enzyme that must be volatile to be removed and non-toxic by drying. Water, ethanol and isopropanol are typical liquids, either alone or in combination. The liquid solution can either be based on aqueous (safer) or solvent. Water mixed in the powders may form bonds between particles of powder, which are strong enough to lock them together. However, the powders may fall apart once the water dries.
Water may not, therefore, be sufficiently strong to build and maintain a bond. A liquid solution that includes a binder is required in such cases. The granulation is milled once the solvent/water has been dried and the powders have developed a more compact layer.
Depending on the characteristics of the powder and the available equipment, the process can be very simple or very complex. The wet mass is pushed through a sieve in the conventional wet granulation process to create wet granules, which are subsequently dried. A subsequent testing process removes granulate agglomerates. Organic solvents are used as an alternative to dry granulation when water-sensitive drugs are processed, or when rapid drying time is needed.
Because for many active substances, direct compression is not the best technology, wet granulation is still a preferred method. Even if the active material is susceptible to hydrolysis, modern equipment (for example, a fluidized bed) prevents all wet granulation issues.
Finally, by adding liquid binders to the powder mixture, wet granulation involves the production of a granule. The single loading and precise feeding of the active pharmaceutical ingredient (API) and a variety of excipients into a continuous blender require both constant, direct compression (CDC) and constant mixing of dry granulation processes.
